2 months ago......replaced all bellows/impeller/gimbal bearing/gaskets/shift cable/pressure tested(the works)...checked alignment(all good)

Last Saturday....took the boat out('86 Wellcraft 23Nova XL/5.7L)....before doing so I checked the outdrive oil......was maybe 1/16" below.....thought all was good......went out/came back had a great time....no problems.....

Back out Sunday....another great day of relaxation.....come Monday morning I see oil/water leaking from the Vent Screw....unscrewed it and maybe less than 1/2 ounce came out........

Question is......Could the water have come from not replacing the seal on the vent screw after checking the level? Sucking in?

You should have a supply of those seals. Every time you unscrew your vent or fill plug you replace them. They are only meant to be tightened down once...

Maybe do a pressure test to be certain?
