I have read the sticky that Don S. has posted for us. thank you Don S.
1)while running these test should the muffs be on??
2)since my boat has set since last year should i spray fog oil or regular oil in spark plug holes first?
3)should coil wire be revoved to prevent start if not useing the muffs?

Fogging oil should have been spayed into the carb during winterization, and you shouldn't need anything in the spring.
As far as starting, if just checking to see if the starter will work, there is no sense in having water too the outdrive. Even if the engine is just cranking over for a few turns.
