Power trim & tilt was working flawlessly until this weekend, when it suddenly quit. I say "intermittently" in the thread title because it worked again for a few minutes while underway then quit again.

I believe the switches are fine because when either the throttle or lower cowl switches are activated there is a "click" in the motor. I tried changing one relay for another (in the fuse panel) with no result.

Is there an area in these engines where the wiring is vulnerable? Or could it be something else?

The relay box in in the lower cowling. The motor is in the transom brackets, Check the routing of the harness from the relay box to the motor. Check grounds to assure they are clean and secure. Check the motor for rust. it may indicate water got in.

Found the culprit but I'm not sure how to fix it. The plug-in electrical connector above the fuse box is loose (or one of the wires inside the connector is loose). Sometimes tugging on a wire or pressing the connector up or down allows the trim to work. I figure adding silicone to the connection would be a bad idea. Any suggestions?

You should be able to remove the wires from the plastic connector. I think they have small fingers holding them in the connector. Now maybe you can adjust the connecting surface with a needle nose plier.
