I have a 4.3l v6 that I have been running, and I have been using ACMR43T plugs in it. However this year we added new cam, rocker arms, intake manifold and a 4 bbl carb. My question is should I run a hotter plug than the 43T, I was thinking 44T or even a 45T.
any suggestions

If you go hotter your will just burn stuff up.
BUT are you running a igntion and/or Coil? It would very much help extra fuel from the new carb burn good, you could run your gap a little bigger on that same plug.... it would help all that stuff

If the camshaft has different specs, it will change the valve timing and final compression of the cyls. You can't tune a carb to compensate for that, if its too far one way or the other.

Assuming you used the same spec camshaft, you should prolly use the same plugs.

-- Joe
