I have never tried pressure testing a drive with it separated. If one did, how do you plug the hole? What is simple and practical and easy to do?

Lowes and/or the Home Depot have the right sized rubber stoppers. Been there, done that. Remember and/or note: (I am assuming you mean how do you seal the oil passage hole, and not the drive shaft hole.....you cant test an Alpha-1 gen II upper by itself. Not the same design (in the driveshaft penetration) as the older Alpha-1's and previous).

Correct.
