Yam. Exciter 370, No Spark In One Cylinder

I have a 99 Yamaha Exciter 370 jet boat. I don't have spark in one cylinder, how can I know for sure if the coil failed or is something else?. Engine works fine on two cylinders now. Also I need to know how to pre mix fuel/oil and what is the ratio?. My oil pumps work fine but I would like to do a pre mix to prevent engine damage if the pump stops working.

Did you ever fix the problem? If so, what was it? I'm having the same issue now.
